Enquanto rapaz, John Nelson Darby estudou em Westminster School e, a desejo de seu pai, a partir de 1815 fez faculdade em Trinity College, Dublin, Irlanda. Por causa da sua consciência, John Nelson Darby renunciou às perspectivas excelentes oferecidas nessa profissão. Já em 1820 entrou em profunda crise interna.
